Terry Pratchett is brilliant and prolific, which makes it hard to know where to start. I've recommended Going Postal as a good introduction to Pratchett. Guards! Guards! may be a better choice if you're already interested in Pratchett and are looking for a good all-around introduction to Discworld. Many of the main characters and concepts (such as L-Space, which I love) are introduced here.
